Gasconade County is located along the south shore of the Missouri River in an area otherwise known as the Missouri Rhineland. Gasconade County covers 526 square miles between the counties of Montgomery, Franklin, Phelps and Osage. The county’s main cities include Gasconade, Owensville, Mount Sterling, and Hermann.

U.S. Route 50, built in 1926, is a major east–west route through the southern portion of the county parallel to Interstate 70. Missouri Routes 19 and 28 have existed since 1922 in Gasconade County, and Missouri Route 100 is adjacent to the historic Lewis and Clark Trail through the northern part of the county.
